Virion Properties

Morphology

Virions consist of a capsid. Virus capsid is not enveloped. Capsid/nucleocapsid is elongated with helical symmetry. The capsid is filamentous, flexuous with a clear modal length with a length of 750 nm. Axial canal is indistinct. Basic helix is obscure.

Electron microscopic preparation and references: Virus preparation contains few virions.

Physicochemical and Physical Properties

The thermal inactivation point (TIP) is at 60-65°C. The longevity in vitro (LIV) is 8-12 days. Although the titer is dependent on the host, the decimal exponent (DEX) of the dilution end point is usually around 4.

Nucleic Acid

The genome is monopartite. Only one particle size of linear, positive-sense, single-stranded RNA is recovered.

The viral genome encodes structural proteins and non-structural proteins.

Lipids are not reported.

The virus does not show serological relationships to potato Y and tobacco wilt viruses.

Under experimental conditions susceptibility to infection by virus is found in several families. Susceptible host species are found in the Family Solanaceae. The following species were susceptible to experimental virus infection: Datura tatula, Hyoscyamus niger, Lycopersicon esculentum, Nicotiana glutinosa, Nicotiana tabacum, Physalis floridana, Solanum jasminoides.

Experimentally infected insusceptible Hosts: Families containing insusceptible hosts: Amaranthaceae, Cucurbitaceae, or Leguminosae-Papilionoideae, Solanaceae. Species inoculated with virus that do not show signs of susceptibility: Cucumis sativus, Gomphrena globosa, Phaseolus vulgaris, Solanum tuberosum.

Diagnostic Hosts

Diagnostic host species and symptoms:

Nicotiana tabacum — vein banding and mosaic.

Nicotiana glutinosa — mottling.

Datura tatula — local lesions and necrosis.

Solanum jasminoides — mottling.

Diagnostic host: insusceptible host species Solanum tuberosum, Cucumis sativus, Phaseolus vulgaris, Gomphrena globosa.

Maintenance and Propagation Hosts

Most commonly used maintenance and propagation host species are Nicotiana tabacum, N. glutinosa.
